Basic Concepts in enzymology, classification of enzymes, cofactors, prosthetic groups, free energy change, transition state and energy of activation, enzyme substrate complex, features of enzyme active sites, effect of various factors on the activity of enzyme pH and temperature Enzyme Kinetics: Simple unireactant system rapid equilibrium approach (Michaelis- Menten equation), The Steady state approach (Briggs and Heldane equation); Heldane relationship between kinetic constant and equilibrium constant, Significance of Km and Vmax values; turn over numbers; ratio of Kcat/Km;Velocity versus substrate curves;multiple substrate reactions, sequential and double displacement reactions; reaction order; 1st order kinetics; zero order kinetics

Course Synopsis

This course is divided into two parts. first part covers basic catalytic features of enzymes including mechanisms used by different enzymes. detail study of catalytical mechanism of three major enzymes (Chymotrypsin, carbonic anhydrase and restriction endonucleases). During second part of this course students will learn about enzyme kinetics, how different biochemical factors effect rate of reaction catalyzed by enzymes. Enzyme inhibitors and types of inhibition.

By the end of this course students will be able to understand 1- catalytic mechanism of various types of enzymes 2- how different factors effect the rate of enzyme catalyzed reaction 3- different moods of enzyme inhibition
